How to Read a Peptide Certificate of Analysis (COA)
Learn how to review a peptide certificate of analysis, including lot numbers, HPLC purity, mass spectrometry, test dates, and traceability.
What a COA is supposed to accomplish
A certificate of analysis summarizes analytical results for a defined material or batch. Its most important function is traceability: the certificate should connect the reported tests to the exact lot used in the laboratory.
A polished document is not automatically a strong COA. Useful certificates contain identifiers, methods, dates, results, and supporting evidence that are internally consistent and specific to the batch.
Start with identity and lot traceability
Before reading the purity number, confirm that the product name and lot number on the certificate match the physical research material. If the sample identifier on an attached chromatogram or spectrum is different, the analytical evidence may not be traceable to the delivered lot.
- Product or compound name.
- Lot or batch number.
- Sample identifier used by the analytical laboratory.
- Date of analysis or certificate issue date.
- Testing organization or laboratory identity when disclosed.
Evaluate the HPLC section
The HPLC section should show more than a percentage. Look for a chromatogram, principal retention time, and a peak table or other result summary. The purity value should be compatible with the displayed integration data.
Remember that chromatographic area percentage is method-dependent. The COA is stronger when the method or analytical conditions are identifiable and the batch-specific chromatogram is available for review.
Evaluate the mass spectrometry section
For peptide identity, mass spectrometry can provide evidence that the observed molecular species is consistent with the expected molecular mass. Review the expected value, the observed signal or assignment, and whether the sample identifier matches the same lot shown elsewhere on the certificate.
Common warning signs
A COA should make verification easier, not force the reader to infer missing information. Treat vague or reusable documentation cautiously.
- No lot number or a lot number that does not match the vial.
- A purity percentage with no chromatogram or peak table.
- An analytical image with an unrelated sample name.
- No test date or no way to connect the result to a specific batch.
- Claims of independent or third-party testing without identifying evidence.
